Artificial thymus tissue enables maturation of immune cells

The thymus plays a key role in the body's immune response. It is here where the T lymphocytes or T cells, a major type of immune defence cells, mature. Different types of T cells, designated to perform specific tasks, arise from progenitor cells that migrate to the thymus from the bone marrow. Researchers at the Max Planck Institute of Immunology and Epigenetics in Freiburg have generated artificial thymus tissue in a mouse embryo to enable the maturation of immune cells. In this process, they discovered which signalling molecules control the maturation of T cells. Their results represent the first step towards the production of artificial thymus glands that could be used to replace or augment the damaged organ.

As part of the immune response, the T cells are responsible for tracking down and destroying intruders and degenerated cells in the body. Their progenitors are formed in the bone marrow. Attracted by chemical signals, they migrate from there to the thymus. This small organ, which is located above the heart, is divided into niches that provide the cells with environmental conditions necessary for their different development phases. The cells mature there into different types of T cells, which are eventually released into the body.

Signalling substances that are active in the various niches of the thymus play a crucial role in the maturation of the progenitor cells. A combination of four proteins ? the two chemokines Ccl25 and Cxcl12, and the cytokine Scf and the Notch ligand DLL4 ? determines whether progenitor cells are attracted to a particular niche in the thymus and how they develop there. It was not previously known which combination of factors is responsible for the development of a particular cell type.

Researchers from the Max Planck Institute in Freiburg have now succeeded in explaining the control mechanism. "The development of the progenitor cells depends on surprisingly few factors and follows simple rules," says Thomas Boehm. "For example, just two factors, namely Cxcl12 and DLL4, are sufficient for the T cell progenitors to complete half of their development. From that point, it takes just a few steps until two types of mature T cells with the CD4 and CD8 surface molecules are formed. Previously, it was assumed that the control process was far more complicated," explains the scientist.

In order to test how the different combinations of the four genetic factors affect the maturation and differentiation of the immune cells, the researchers created an artificial thymus environment in the mouse embryo. To do this, they switched off the transcription factor Foxn1. Fox1 ensures that all of the genes in the DNA that code for the four different proteins are read. Foxn1 is thus a kind of "general switch" with which all of the relevant genes can be turned off all at once. Having generated non-functional thymus tissue in this way, the researchers then activated the genes again individually and in combination and observed the associated effects on the development of the progenitor cells.

Interestingly, the scientists were also able to attract the progenitors of B lymphocytes and of mast cells to the artificial thymus environment and allow them to mature there. These two types of immune cells normally only develop in the bone marrow. By switching off the Notch ligand DLL4, the researchers were also able to control the maturation of these cells in the artificial thymus. This would suggest that the genetic factors at work in the bone marrow are similar to those in the thymus.

"Our results are not only of relevance to the understanding of fundamental immunological processes," says Thomas Boehm. "They also constitute early milestones in the development of artificial thymuses. This means that one day we may be able to help patients whose thymus has been damaged, for example as a result of cancer treatment," hopes the scientist.

US stocks fall on worries about Europe's economy

(AP) ? Stocks fell Thursday for the third day in a row after reminders that Europe has not solved its debt crisis and the U.S. economy is far from healed, despite progress on both fronts.

The Dow Jones industrial average fell 58 points in early trading to 13,069. The broader Standard & Poor's 500 fell nine points to 1,397. The Nasdaq composite index fell 15 to 3,090.

The declines came despite incrementally good news about the U.S. economy. The government said that the number of people seeking unemployment benefits last week dropped by 5,000 to about 359,000, the lowest since April 2008

The government also said that the U.S. economy grew at an annual rate of 3 percent in the fourth quarter, which met analysts' expectations and was the fastest pace since spring 2010.

But there was reason for concern, too: The government also said that many more people than it originally estimated filed unemployment claims in recent months. And economists believe the country's growth has slowed to about 1.5 percent.

European markets fell across the board, despite a report from Germany that its unemployment rate fell slightly over the month. The benchmark stock index was down 1.9 percent in Germany and 1.3 percent apiece in France and Britain.

Though Greece is no longer on the brink of default, deep problems remain for the continent, where stronger countries are arguing that they shouldn't have to bail out weaker ones.

In Spain, workers took to the streets to protest government spending cuts, underscoring the difficulty that governments will have in reining in overblown spending.

The price of oil fell 65 cents per barrel in New York to $104.73. France's prime minister said there's a "good chance" the U.S. and Europe will release oil reserves, which could drive down prices by adding supply.

President Barack Obama was expected later Thursday to call on Congress to end tax breaks for oil companies. The price of oil has doubled since October, and the price of gasoline has risen about 20 percent this year.

Among the stocks making big moves:

? Illumina, a diagnostics company, rose more than 4 percent to $52.06 after the Swiss pharmaceutical giant Roche Group raised its offer for the company to $51 per share.

? Best Buy fell more than 6 percent after announcing plans for big spending cuts, including closing 50 of its big-box stores.

? JetBlue fell 8 percent, two days after passengers said a pilot came unhinged and forced a JetBlue flight to make an emergency landing on a flight from New York to Las Vegas.

Solar storm seen from inside and outside Earth's magnetosphere

ScienceDaily (Mar. 28, 2012) ? For the first time, instrumentation aboard two NASA missions operating from complementary vantage points watched as a powerful solar storm spewed a two million-mile-per-hour stream of charged particles and interacted with the invisible magnetic field surrounding Earth, according to a paper published March 28 in the Journal of Geophysical Research.

The spacecraft, NASA's Two Wide-angle Imaging Neutral-atom Spectrometers (TWINS) and Interstellar Boundary Explorer (IBEX), observed the impact from inside and outside Earth's magnetosphere, respectively. The energetic neutral atom (ENA) cameras aboard each spacecraft enabled global imaging of the magnetosphere, the invisible bubble that protects Earth from the majority of charged particles from the Sun, as it compressed in response to sharply faster solar wind.

The storm, observed April 5, 2010, also is thought to have caused an important communications satellite, Galaxy-15, to founder and drift, taking almost a year to return to its station.

"Many satellites above Earth are in geosynchronous orbit. Like heavy traffic on a Los Angeles freeway, they have to stay in their lanes because they could, theoretically, collide," says Dr. David McComas, assistant vice president of the Space Science and Engineering Division at Southwest Research Institute, and principal investigator of the IBEX and TWINS missions. "More likely, however, is that they will get too close together and their radio frequencies will interfere with the operation of nearby satellites, which could hamper activities on Earth."

The IBEX images (taken from a distance of around 180,000 miles) show an immediate compression of the magnetosphere as it was impacted by charged particles from the solar wind. ENA global imaging enabled the team to determine the precise timing of the compression, to within ?9 seconds. Minutes later, one of the TWINS spacecraft, carrying identical ENA sensors that provide stereoscopic imaging, observed changes in the inner magnetosphere (from a much-closer 28,000 miles). A magnetospheric structure called the "ring current" traps charged particles that gyrate around magnetic field lines. About 15 minutes after impact, the trapped particles propagated down the field lines toward the poles and into Earth's atmosphere, where they produced additional ENAs. The brief time delay in losing particles to the atmosphere suggests that internal magnetospheric processes take some time after compression from the initial impact.

"This imaging gives us a better global picture of the evolution of the magnetosphere -- especially of the processes by which the Sun injects energy into the magnetosphere -- than has ever been available before," says McComas. "It's a cool combination of two data sets that opens the door to much more sophisticated global studies."

To produce the new images, the team also developed a series of techniques to process the imaging data, including improved procedures for binning, differential background subtraction and "statistical smoothing" of images.

"Understanding how solar events develop and impact satellites is like understanding the processes that cause extreme weather events on Earth to develop and destroy homes and businesses," says McComas. "Engineers use weather data to know where and how they need to strengthen buildings against various types of weather threats. The more we know about the processes occurring in space, the better engineers can design satellites to protect them from space weather hazards, which is increasingly important in our highly technological world."

SE Pa. church subjects youth group to mock kidnap, interrogation; outraged mom files complaint

PITTSBURGH - A southeastern Pennsylvania church subjected members of a youth group to a mock kidnapping and interrogations without telling them it was staged, and the outraged mother of one 14-year-old girl has filed a complaint with police.

The pastor of the Glad Tidings Assembly of God in Middletown said the church is "so saddened" that the girl was traumatized at the Wednesday evening youth meeting.

But the pastor, John Lanza, said Friday there have been emails of support from other students at the church, about 10 miles southeast of Harrisburg, because the intent was to prepare them for what they might encounter as missionaries. He didn't disclose the names of those involved but said the mock kidnappers included an off-duty police officer and a retired Army captain.

"It was a youth event, to illustrate what others have encountered on a regular basis," he said, adding that the focus of the lesson was "the persecuted church" in other countries.

Lanza said there were about 17 students at the meeting and the mock kidnappers covered the students' heads, put them in a van and interrogated them. Neither the students nor their parents were told about the raid beforehand, he said, though it was discussed with the parents of one youth who might have health issues.

TV station WHTM interviewed the girl who complained.

"They pulled my chair out from underneath me, and then they told me to get on the ground," she told the station. "I had my hands behind my back. They said, `Just do as I say, and you won't be hurt.'"

The girl said the teens were taken to the pastor's house, where it appeared he was being assaulted. Eventually, she said, the adults in charge revealed it was a staged event.

"They heard me crying," the girl said. "Why not right then and there tell us it was a joke, when you see me crying?"

Lanza said the church has conducted similar events at least twice before, adding that "there was much thought given to the safety aspect."

"If anyone was ever uncomfortable, they would be removed" from the exercise, Lanza said, though part of the idea was to shock the students with the experience.

Lower Swatara Township police Chief Richard Wiley declined to comment until an investigation into the raid is complete. The names of the mother and daughter who complained haven't been made public.

There could be consequences if the teens didn't know what was going to happen and didn't agree to be a part of the event, Dauphin County First Deputy District Attorney Fran Chardo said.

"It's extremely disturbing," Chardo told WHTM.

Tom Copeland, who studies international affairs and terrorism at Geneva College, a Beaver Falls school that emphasizes a "Christian worldview," questioned the wisdom of submitting a youth group to a mock kidnapping.

"It just seems inappropriate for that age group. You would think there would be permission from the parents," Copeland said, adding that he's never heard of anything like that happening at a church.

He said that while there have been highly publicized episodes of violence directed against Christian missionaries in other countries, it seems those countries' local religious groups are more at risk.

One security expert said role-playing training is sometimes conducted "at the quasi-military level" for groups that are going to work in war zones.

Daniel Karson, chairman of business intelligence and investigations at Kroll, a worldwide risk consulting company with headquarters in New York, said the idea of conflict zone training "is to acclimate someone to a possible situation that might arise." The training might involve everything from what items they should take to the country to a review of who they're meeting and security conditions there, he said.

Lanza said members of the church have made numerous mission trips overseas and have learned to be cautious. He said they were planning a trip to Mexico but reviewed current news and advisories and decided it was unsafe.

Lanza said he "would love to" apologize to the girl and her mother but feels he can't until the police investigation is done. He said the church wants to keep doing the program but would make changes.

"I would find a way that we could continue to keep the shock value," he said, "but I would find a way to inform the parents (beforehand)."

Scientists have debated whether climate change or human activity wiped out the world's megafauna. In Australia new evidence points to hunting, and only hunting

Around 40,000 years ago, the giant kangaroo disappeared from Australia. So did Diprotodon (rhinoceros-size wombats) and Palorchestes (tapirlike marsupials) as well as supersize birds, reptiles and some 50 other so-called megafauna?big animals. And now a record of fungal spores pulled from the swamp at Lynch's Crater in the northeastern corner of the continent reveals humans as the culprit.

"The megafauna declined soon after the time that we know people arrived in the region," explains zoologist Christopher Johnson of the University of Tasmania, lead author of the report published March 23 in Science. "We conclude that humans, not climate, caused the extinction."

The basis for the charge rests on two mud cores?one stretching from around 130,000 years ago to 24,000 years ago and the other ranging from roughly 50,000 years ago 3,000 years ago. The long core reveals how the local environment reacted to two previous periods of cooling and drying. Judging by the Sporormiella fungus?which only releases its spores when in the dung of plant-eating animals?big animals fared well during these climatic changes, munching happily on the changing vegetation of the area.

That changed roughly 41,000 years ago, when the number of spores in the core "dropped almost to zero," the researchers wrote. The record in mud also notes an increase in charcoal from large-scale fires and new types of vegetation?eucalyptus trees and the like with grasses beneath, much as seen today. A shift in climate could alter plant populations, but this vegetation change actually precedes by roughly 10,000 years the most recent climate shift within the core's age, to cooler and drier conditions. "Climate change played no role in megafaunal extinction in Australia," Johnson concludes.

Using the second core, the researchers then focused on the period between 43,000 years and 38,000 years ago to try to understand how humans eliminated the megafauna. Humans could have hunted them to extinction or set fires that changed the landscape so much that large herbivores could not survive. But examining the core in 100-year increments showed that the shifts in vegetation happened after the near disappearance of Sporormiella spores?not before as would have been expected if fire-triggered vegetation changes had caused the extinctions. "It could not have been vegetation change due to firing of the landscape, as other people have proposed, because those things followed megafaunal decline," Johnson argues.

In fact, the disappearance of the big plant-eaters seems to have set the stage for fires, allowing the buildup of the dry grasses and other fine fuels that spur burning like the catastrophic wildfires still seen in Australia today. At Lynch's Crater, the disappearance of the large plant-eaters saw an increase in grasses within 300 years, then acacias, eucalyptuses and other hard-leaved plants within 400 years, and, ultimately, a rise in the pollen from forest trees some 1,600 years later. Even today, many of the plants still extant in Australia boast features such as protective spines that would discourage grazing by megafauna or big fruit and seeds that could only be dispersed by large animals that no longer exist?a landscape shaped by ghosts. "These plants are now anachronistic," Johnson observes.

The findings seem to close the case against modern human hunters, although they remain to be confirmed at other sites throughout the continent. And, on every continent except Africa, human arrival and large animal extinctions seem to coincide, so the case may also extend globally. (The reason large animals did not vanish in Africa is perhaps because they co-evolved with us and learned to be wary of this stalking, hairless, upright ape.)

An analysis of ancient DNA from extinct species such as woolly mammoths and rhinoceroses suggests that "climate has been a major driver of population change over the past 50,000 years," according to a 2011 Nature paper that also suggests climate and humans may have colluded to push such species to extinction. (Scientific American is part of Nature Publishing Group.) Johnson, for one, isn't sure climate should be blamed. "Extinctions in other parts of the world were remarkably similar in pattern and severity to those that occurred in Australia," he notes. After all, human hunting wiped out similar species from in neighboring New Zealand less than a millennia ago. "This seems to strengthen the view that human impact?mainly hunting?was the predominant cause in other places as well as in Australia."

As for the specific weapons and techniques, they remain unknown. "It is almost certain that hunting of large animals was one of the things [early Australians] did, but we'll probably never know exactly how they did it," Johnson says. In this case, it wasn't the weapons that fingered the perpetrators but an absence of fungal spores that reveals an ancient human deed.

(AP) ? India's Tech Mahindra said Wednesday it will take full ownership of its Satyam subsidiary, creating India's fifth largest outsourcing company.

Satyam Computer Services nearly collapsed in one of the largest frauds in Indian corporate history in 2009.

Three months after Satyam founder Ramalinga Raju confessed to inflating company assets by $1.5 billion, Tech Mahindra bought a controlling stake in the company and renamed it Mahindra Satyam.

The boards approved the long-anticipated deal Wednesday. The enlarged company will have revenue of $2.4 billion and over 75,000 employees serving 350 clients in 54 countries.

Investors will get two shares of Tech Mahindra for every 17 shares of Mahindra Satyam.

"The Mahindra Satyam turnaround is a shining story of determination and grit and now comes to its most important chapter, with this merger," Mahindra Satyam chief executive C.P. Gurnani said in a statement.

Tech Mahindra is owned by India's Mahindra & Mahindra Ltd. and British Telecommunications. The Mahindra Group will own 26.3 percent in the combined entity and British Telecom will own 12.8 percent.

The takeover requires shareholder and regulatory approval.

IIFL Capital analyst Sandeep Muthangi said the takeover would result in cost savings and better competitiveness.

"It's favorable for both companies," he said, adding that India's IT sector will likely see more consolidation in the future.

Shares in Tech Mahindra were up 3.7 percent and shares in Satyam were up 4.7 percent in midafternoon trade in Mumbai, beating the benchmark Sensex index's 1.2 percent rise.

Baylor has made remarkable rise of redemption

WACO, Texas (AP) ? Buddy Jones wakes up almost every morning singing "Amazing Grace." He can't help himself.

Jones, the board chairman of the Baylor University regents, is in constant awe of all the great things his school is experiencing these days.

Only a few months after quarterback Robert Griffin III became Baylor's first Heisman Trophy winner, the men's basketball team is in the NCAA round of 16 for the second time in three years. So are coach Kim Mulkey's top-ranked Lady Bears, undefeated and four wins from a national championship that would make them the first 40-win team in the history of NCAA Division I basketball.

"Baptists talk a lot about redemption and Baylor University was redeemed two summers in a row, from what looked to me to be a surefire, at least near-death experience, maybe death completely," Jones said. "Because of hard work, and good fortune and the way things worked out, we were given a stay of execution and we hopefully have begun making the most of it."

If that sounds like overstating things, consider the facts.

Jones and other Baylor officials spent much of their last two summers lobbying just to make sure the Big 12's smallest and only private university didn't find itself without a spot in a major athletic conference ? a situation that would have been devastating not just to prestige but to the bottom line. The league was teetering on the brink of collapse amid speculation of potential super conferences and the departure of four schools.

Yet, the story goes much deeper for the world's largest Baptist university, making the culmination of all this success even more remarkable.

"It's almost of epic proportion," said Bob Beaudine, whose executive search firm brought men's basketball coach Scott Drew and athletic director Ian McCaw to Baylor nearly nine years ago in the aftermath of a tragic scandal that shook college sports like nothing before.

In the summer of 2003, basketball player Patrick Dennehy was found shot to death after he had been missing for six weeks. Teammate Carlton Dotson pleaded guilty to murder. The ensuing investigation uncovered NCAA violations, illegal tuition payments and unreported failed drug tests that led to the resignation of coach Dave Bliss, who was secretly recorded by an assistant coach of trying to persuade others to cover up misdeeds by portraying Dennehy as a drug dealer.

Drew inherited a roster decimated by the departure of the three top scorers, who left under relaxed transfer rules. There were school-imposed sanctions reducing scholarships and recruiting visits, and the NCAA limited the Bears to playing only conference games in his third season.

With three NCAA tournament appearances the past five years, an NIT championship game in that span and already a school-record 29 wins this season, many of those bad memories are quickly fading away. The Bears play Xavier on Friday night in Atlanta.

"You don't hear Baylor and think of that whole episode anymore. You think of RG3, Brittney Griner and this guy right here (potential NBA lottery pick Perry Jones III)," senior forward Quincy Acy said. "It's going to take time to completely erase it and for us to become a complete powerhouse, but I think we're headed in the right direction."

Ah, yes. Griffin and Griner, the 6-foot-8 junior phenom.

Griner, the only women's college player who is a U.S. Olympic team finalist, helped lead the Lady Bears to the NCAA Final Four as a freshman and a regional final last year before losing to Texas A&M, the eventual national champion they had already beaten three times.

"It's just a great time to be a Bear right now," said Griner, who became only the second woman to dunk in an NCAA game during Tuesday night's win over Florida. "On campus, everybody is really excited. The community, fellow classmates, everybody's behind us."

All of Baylor's sports teams, men and women, have qualified for the postseason so far this academic year, and the 400-plus athletes on campus had a combined 3.16 GPA last fall.

Griffin, who got his political science degree in three years, set or tied 54 school records in 41 games and then went out and charmed everyone at the NFL scouting combine without throwing a pass. The dual-threat quarterback led the Bears to only the second 10-win season in their 112 years of football. They had never even had a winning record in the Big 12 before RG3 arrived with coach Art Briles.

The Washington Redskins traded three first-round picks and a second-rounder to St. Louis for the No. 2 overall pick in next month's NFL draft. That's presumably to select Griffin since the Indianapolis Colts, now without Peyton Manning, are expected to take Heisman runner-up Andrew Luck from Stanford first overall.

Baylor President Ken Starr ? yes, the former independent prosecutor best known for his investigation into the Clinton White House and the Monica Lewinsky scandal ? believes athletics provide a voice through which the university speaks to the entire world.

"I'm so proud of the way our athletes use their God-given gifts coupled with their very hard work to bring great joy and pride to all of Baylor nation," Starr said. "The voice of Baylor athletics is quite eloquent right now."

While hard to determine the true impact of athletic success on freshman applications, the numbers have drastically increased.

There are more than 40,000 applications for the upcoming fall semester for only about 3,000 freshman spots. That's up from 15,458 applicants for the Fall 2005 class, right after the Lady Bears won their first national title. Average SAT and ACT scores for incoming freshmen also have significantly increased during that time.

"I think our national championship erased a lot of negative publicity from what had happened with men's basketball," Mulkey said. "I think timing in all of our lives is so important ? the timing of me leaving Louisiana Tech to go to Baylor, the timing of when we won the national championship, the timing of the Heisman Trophy and men's basketball doing well and women's basketball."

More than $250 million in new athletic and academic facilities have been added in the past decade to the campus that's home to about 15,000 students in Central Texas. Another $120 million in capital improvements are under way. The Bear Foundation, the primary fundraising arm supporting the school's 19 athletic programs, contributed nearly $7.7 million for scholarship support during the 2010-11 academic year. It was the seventh consecutive year with a record total.

There are also plans for a new campus football stadium, just off Interstate 35 along the banks of the Brazos River in the city of about 120,000 people. The family of former Houston Astros owner Drayton McLane Jr., a 1958 Baylor grad and former regents chairman, recently gave the school the largest capital gift ever ? more than $20 million ? for the stadium the school hopes to have for the 2014 season.

"(The stadium) has been the talk of the town. Robert Griffin had a great year, and that got things rolling, and the basketball is continuing. You need good news, and this is good economic news for Waco," said Sammy Citrano, owner of the two George's restaurants known for their big chicken fried steaks. "I've been here 26 years and this is a great feeling."

Bliss has since shared his testimony and counseled young coaches, telling them he allowed the competitive world of college athletics to compromise his beliefs. He still gets calls with questions about what happened, usually when the Bears are having the kind of success that puts that dreadful time even further behind them.

"I try to make it as positive for Baylor as I can, because I put them through enough," said Bliss, now coach, athletic director and dean of students at Allen Academy, which serves nearly 300 students in kindergarten through 12th grade near Texas A&M.

"I chose to go to Baylor because I wanted to help be part of the solution, and the aspect of what they have accomplished doesn't surprise me because I think I saw a lot of the great things that were possible there," Bliss said. "I'm obviously thrilled for what they're accomplishing, and the way they're accomplishing it."

(AP) ? A 101-year-old Utah woman who ushered in her birthday with a tandem paraglide ride last year soared into the record books Tuesday.

Great-great grandmother Mary Hardison, of Ogden, has been officially recognized as the "Oldest Female to Paraglide Tandem" by Guinness World Records. She has supplanted a 100-year-old woman from Cyprus who took her flight in 2007.

Hardison flew with an instructor on Sept. 1, 2011, while four generations of her family watched and cheered. The flight went smoothly and even included a few tricks that Hardison encouraged.

Hardison said she wanted to paraglide because her 75-year-old son began doing it as a hobby.

"I didn't want him to get too far ahead of me, so I decided that I'd go too," she said Tuesday.

Fear never entered the equation for Hardison, who previously rode all of the adult rides at Disneyland to celebrate her 90th birthday.

Hardison said she was shocked to learn her flight had become a world record.

"To me, I was just going on a little ride," she said. "I had no idea I'd break a record."

Hardison said her active lifestyle has already inspired others her age to pursue similar adventures.

"Some people have said, 'Well, if you can do that at your age then there's no need for me to just sit around,'" she said. "My desire is for the elderly to keep on going. Do things as long as you are physically able. Be positive. Friends don't like a grumpy person."

When Hardison decided to take the birthday leap, family members were concerned for her safety. After all, it was her first flight and she was the oldest customer the paragliding company had ever taken into the air.

"My first reaction was she was nuts," said daughter-in-law Bonnie Hardison. "But everyone is just so excited and thinks this record is great."

As for any future flights, Mary Hardison, who spends her days quilting for charities, is just playing it by ear.

"If the notion strikes, I'll go again," she said.

The rest of the prices are as follows: $9.95 per month for 500MB, $19.95 for 1GB, $34.95 for 2GB, and $49.95 for 4GB. That $10-20 range is where the NetZero HotSpot shines. The only comparable plan in that price range is from Virgin Mobile, which offers 500MB of data for $20 per month?double the price of NetZero. So if all you need is light email and Web access, NetZero's HotSpot is a good deal indeed.

But for the same $34.99 that gets you 2GB on NetZero, the Clear Spot 4G Apollo?($99.99, 4 stars) gets you unlimited data, albeit at capped speeds. But $49.99 gets you unlimited data from both Clear and Virgin Mobile, with no preset speed caps. Even Sprint offers 3GB of 3G or 4G data for $34.99 per month, 6GB for $49.99, or 12GB for $79.99, though those plans require a contract. So the larger your data needs are, the better off you are going with a different carrier. Additionally, NetZero offers its broadband through Clear, which is only available in a limited number of areas, so make sure you're covered before buying in.

As far as I can tell, the NetZero 4G HotSpot is the same exact device as the Clear Spot 4G Apollo, only with a different logo. Physically, the HotSpot is a big black box, measuring 3.4 by 3.4 by .7 inches (HWD) and 4.4 ounces. It probably won't fit into your pocket, but it'll easily slip into your bag. On the plus side, all of that space allows for a large 2200 mAh battery, which makes for some excellent battery life. I was able to get 6 hours and 23 minutes of solid 4G streaming on one charge.

The HotSpot has just two buttons: a Power button on the left, and a Mute button on the right. They're easy to mistake for one another, and the Mute button isn't necessary; that function could have been included on the Web-based management console. There's also an external antenna port on the bottom of the device for boosted reception, though I got a good signal without one. The best feature is the 2-inch LCD on the front of the HotSpot. It tells you the device's battery life, amount of data transferred, number of devices attached, password, signal strength, SSID, and the time connected. It's a lot of information, and it's all large, clear, and easy to read.

Setup and Administration
To connect, simply turn the HotSpot on, and use your computer or device's wireless connection manager to connect to the HotSpot using the SSID and password displayed on its screen. Since this is a mobile hotspot, you don't need any drivers for it. You can connect up to eight devices this way; I connected three at once, and they worked fine. You can also tether the modem to a computer via USB. I tethered the HotSpot to a laptop running Windows 7 and got speeds that were about on par with Wi-Fi. You can also connect to the device while it's charging, which is a feature many hotspots lack.

There's a nice Web-based management console, which you can access by sending a connected device to http://netzerohotspot. From there, you can monitor signal strength and battery life, check out who's connected to the router, and control a wide range of router settings. It's fairly comprehensive, allowing you to change the screen backlight, set your Wi-Fi options, enable port forwarding and firewalls, and modify a host of security options. There's also a neat feature that lets you control the speed at which your data is used. Choosing Lightspeed caps your downloads to 1Mbps, while Warpspeed allows you to reach 10Mbps. Free and $9.95 plans are already set to Lightspeed, while plans that cost $19.95 are automatically set to Warpspeed. These settings can be changed at any time, and are a good way to help conserve precious data.

Performance and Conclusions
I tested the HotSpot in New York City. We didn't have another WiMAX hotspot on hand to test it against, but since this is the same hardware as the Clear Spot 4G Apollo, I expect it to perform similarly. That's a good thing, since the Apollo outperformed a Novatel Wireless MiFi 3G/4G Mobile Hotspot 4082 from Sprint (Free, 4 stars) when I tested it.

In our?21-city tests?last year, we got 3Mbps average download speeds on Sprint's 4G network, which is the same as Clear's.?For downloads, the HotSpot averaged a solid 4.5Mbps down. Upload speeds weren't nearly as fast, at an average of .7Mbps up, but those speeds are near identical to what we saw on the Apollo. And either way, these are respectable numbers, especially when there's a data limit in place?you don't want to eat your way through a month's worth of data in 90 minutes.?

According to NetZero, the HotSpot should work within a 150-foot range. Speeds held up within at least 50 feet of the device, then started to drop off, which is common for most hotspots I've tested. It can still be used at 150 feet of distance, but speeds will be painfully slow.

The NetZero 4G Hotspot is a good choice for a very niche group of users. If all you need is 1GB or less of mobile broadband per month, the 4G HotSpot can't be beat. But if your data needs run deeper, you're better off looking elsewhere. Clear and Virgin Mobile are both good places to start. Both carriers offer unlimited data for $50 per month, which means you can even use those hotspots as a primary home internet connection.?

D'Arcy qualifies for Olympics, 4 years after ban

(AP) ? A reckless celebration after the 2008 Australian selection trials cost Nick D'Arcy his spot at the Beijing Olympics, resulted in a criminal conviction after a barroom altercation with another swimmer and damaged his reputation.

After dominating the 200-meter butterfly on Sunday night to gain selection for the London Olympics, he planned a quiet dinner with his parents ? and maybe have a few friends over when he gets home later in the week.

"I'll keep it a little bit quiet," the 24-year-old D'Arcy said after winning the 200 fly in 1 minute, 54.71 seconds, ahead of second-place Chris Wright in 1:56.40.

After being barred from competing at Beijing and the 2009 world championships in Rome, he is not taking any chances ahead of his Olympic debut and a chance to take on American Michael Phelps.

D'Arcy was only recently cleared to be eligible for selection at London by the Australian Olympic Committee following protracted legal proceedings triggered by an altercation at a Sydney bar with former Commonwealth Games medalist Simon Cowley after Australia's previous Olympic trials.

He was convicted of recklessly causing grievous bodily harm to Cowley, who sustained fractures to his jaw, eye socket, cheekbone and nose. But his sentence of 14 months, 12 days in jail was suspended, meaning he did not have to serve time in prison unless he breached certain conditions.

He also lost a civil case against Cowley and was ordered to pay damages, but filed for bankruptcy last November ? angering his critics again.

D'Arcy said his focus is now just "staying on the right path".

"It has affected me, it's impossible to argue otherwise," he said. "It has been a massive journey, it has been a public journey. Performance-wise in the pool, and out of the pool, it has been a very challenging four years.

"I think that is something that is going to make me a better athlete, just because I have had to show that mental discipline ? especially leading into events when there is other things hanging over my head."

D'Arcy knows that people have different opinions of him, but he wants to channel his energy into competition: "I don't find a lot of positives from hanging onto the past."

"How I'm perceived in the public is not something I have control over," D'Arcy said. "It would be great to have everyone behind me, but I'm realistic.

"I've had some ups and downs. It's just great to come out on top. It has been a rough ride at times. I think I've come out of it a better athlete for it."

D'Arcy said he considered quitting at times, but his family, friends and a rare win over Phelps in a minor meet at Santa Clara last year kept him going.

"There have been some times when I wasn't sure if I was going to continue on, but days like today make it all worth it," he said. "It's a fantastic feeling to be able to go overseas and represent your country as part of the Australian swim team. I'm just honored to be a part of it."

Triple Olympic gold medalist Stephanie Rice won the 200 individual medley Sunday and will defend her title in London.

She won the 400 IM last week. Despite persistent pain in her right shoulder since surgery last year, she held off world championship silver medalist Alicia Coutts to win the 200 IM in 2 minutes, 9.38 seconds ? the fastest time in the world this year.

It was also the fastest time Rice has swum without the aid of the high-tech suits that are now banned in competition.

"The preparation has been so rough and it has been hard for me mentally to be confident," she said. "I'm really happy to have come away with a PB in these suits and, more than anything, just to execute the race I wanted to execute."
